I have a 3TB external drive connected to my Mac. It contains 2.5TB of data. If I use Finder > Encrpyt to encrypt the drive, how long will the encryption take? A few minutes or multiple hours? If it has to somehow read and re-write all the data, my guess is it’ll probably take about 20 hours with a FireWire 800 connection.

I wouldn't argue with that estimate. If I had to do it, I would at least kick it off before going to bed. But more likely I would kick it off before a weekend or something like that.
If you decide to go ahead with this, report back and let us know how long it took.
