It is just a matter of setting it up. Without more details, I'll have to guess at what you have set up already. So I'll assume your iMac is set to your Apple ID. And then your wife's iPhone is set to her Apple ID.
On her iPhone, you would check to make sure she is signed into iCloud in the Settings app. Also, check to make sure she hasn't turned off location services. Look in Settings, Privacy and make sure it is on. Then look further down and make sure it is enabled for Find My Friends.
Then have her go to the Find My Friends app. Tap the add button. Then enter your Apple ID email address. Choose Share Indefinitely.
That should be it. Now if you use the Find My Friends from iCloud.com or using Notifications Center on your iMac, you should see the location of your wife's iPhone.
