My wife’s name in Contacts is listed first name-last name. Settings are Sort last-first, and show as first-last. I have tried reversing these and making them the same, but the result is Siri always asks “Which one, and gives me both versions to choose from.” since I communicate often with my wife, this is frustrating and wastes time. Anything I can do to fix this?

Make sure you don't have two contacts with her name. Look alphabetically for first+last and last+first. For instance, if her name is Jane Doe, look for Jane Doe and Doe Jane in the contacts app. So look under "D" and "J." I'll bet you have her there twice.
I had checked for that earlier and there's only one entry in Contacts. Update: I seemed to have solved it by putting a phonetic field in for her first name "Kelly." Now I just get one name and Siri's ready to proceed. Go figure. Thanks.
I don't have an iPhone 4S but this ought to work based on other people's reported experiences:
Assign a "nickname" to your wife's entry in Contacts. "My Wife" should do, "Hunny-Bunny" might work, or you could try "Kelly" there.

Look under your own name in contacts and see if you listed your wife as your spouse, that may be why. If so, just delete her as your spouse in your contact info...should solve the problem!
Remove your spouse with the Delete key? If only it were that easy....
