Depends on the video, its location and permissions. Some videos, like the ones I publish from MacMost, have a podcast version you can download as an .mp4 and use. But others are only in Flash format on sites like YouTube. In many cases the video creator has done this on purpose so that the video cannot be freely taken and used without permission.
If you don't see a download link of some sort, it couldn't hurt to email the video creator and ask them.
