How can I use highlight selection markup tool in the mark up toolbar in quick look. Highlight selection markup tool is next to the text insert tool in the markup toolbar. Even while viewing a PDF in quick look it does nothing. Does it work similar to highlighting text in the preview app, if yes then how??

Which markup tools work depends on what type of file you have selected. I tried the highlight tool with a PDF and it wasn't selectable. But there are many types of PDFs, so perhaps it only works in some cases.
But obviously it isn't working for you with the PDF you want to highlight. So I would just open the PDF in Preview and highlight it there.
